Lysis Buffer, for resuspension of the cell, what exactly should i put in, because my doc said NaCl & Phosphate

To get water to move into the cell you use the hypotonic solution.KCl. Which may eventually get them to lyse not immediately though. If you want to look at specific cells, this will not be helpful because all of your cells will be gone. Looking at white blood cells...use a little Methanol:acetic acid solution after 10 min. Which will lyse the RBC's immediately leaving the WBC's and have nice cells to look at.
